In memory of Urs Graf’ (1972/1975) is Flavin’s illuminating installation with pink, yellow, green and blue fluorescent light in the four corners of the front court and arcade of the Kunstmuseum Basel. 💡💡💡❌💡

Clearing the opaque and coloring the clear
“Tissue clearing” is a technique that facilitates 3D observation in opaque biological tissue and even whole organs. Through a series of chemical treatments, washes, and final mounting solution, samples become optically transparent. This reduces the need for cutting thin slices normally required for microscopy (swap right to see a cleared mouse brain).
Many tissue clearing methods were published in the last decade. Some methods use organic solvents to render tissues transparent. Newer methods are strictly aqueous, which benefits both the preservation of delicate proteins and user safety. The work shown here demonstrates a set of simplified protocols optimized for quality and practical execution (currently unpublished).
This image shows a ~1.4mm thick coronal slice through the middle of a mouse brain. A Zeiss LSM800 confocal microscope with 10x water immersion objective was used. The two channel, tiled, z-stack scan required about 3 hours to complete.
Thy1-YFP-H (green) is a random insertion BAC transgene expressing fluorescent protein very strongly in an isolated variety of excitatory neurons. Pyramidal cells of the neocortex form the upper semi-circle and many axonal projections that crisscross the interior. .
Vip-Cre; Ai14 (magenta) is a pair of targeted insertion transgenic alleles that result in expression of fluorescent protein (tdTomato) in the subset of inhibitory neurons that are vasoactive intestinal peptide positive.

More light emmision compounds!! From left to right: Rhodamine B, Rubrene, Rhodamine 6G; 9,10-Bis(Phenylethinyl)antracene and 9,10-Diphenylantracene, all of them in ethylphthalate as solvent. Product of Wittig reaction under 365nm UV light. The reaction was carried out between two equivalents of benzaldehyde and the Wittig ylide. This last one was made by treating α,α'-Dichloro-p-xilene, a halogen derivate with two equivalents of triphenylphosphine. The final product ( 1,4-Bis[2-cyano-2-phenylvinyl]benzene ) has a strong emission peak in the blue zone of the electromagnetic spectrum. Such a fun project!! It is amazing how two diferent colourless compounds, when they condense, they increase the amount of molecular orbitals so that it then absorbs in the visible spectrum and develop fluorescence properties.
