Lab scissors continued --- Dr. Mary McMillan
In my last post I showed you my lab scissors – better known in the business as restriction enzymes.
So I thought I should follow up and show you what they do!
This is a picture of an agarose gel – used to separate fragments of DNA according to their size. Each glowing band on the gel is a group of DNA molecules all of the same size. In the lane on the left I have loaded a DNA ladder, which is a collection of DNA fragments of known sizes. We can use this like a ruler to measure the size of other fragments of DNA on the gel.
In the middle lane I have loaded one of my DNA fragments that hasn’t been exposed to my restriction enzyme. In the lane on the right I have the same DNA sample, except I have mixed the DNA with one of my restriction enzymes. The enzyme has cleaved the DNA, resulting in shorter fragments.
My main use for this technique is to look for genetic variation within certain regions of DNA. If there are changes to DNA, it can change the restriction sites, or places where the enzymes will cut DNA. Sometimes it will mean there is no longer one of these sites, and the DNA will remain uncut. Other times changes to DNA can change where the restriction sites are, moving them closer together or further apart, so that I get different size fragments after the digestion. By looking at the different patterns of bands on a gel like this I can figure out if different people have the same version, or different versions of a gene. So my lab “scissors” are pretty useful!

Children & emotions 👶 With every client, I’m reminded of the fragility of childhood and the imprints formed early on. The messages picked up and the interpretations made “I’m not good enough”, “I must do better”... Something I often witness is adults aiming to shut off a child’s distress with comments such as “don’t be so silly”, “come on, don’t be a baby now” or with simple distraction methods “would you like a biscuit?”. I’m also guilty of the above, as sometimes it’s hard to take care of others’ emotions when you’re pretty frazzled yourself! But the messages coming through as a result of these quietening methods are judgement, children who feel their emotions aren’t validated, that their experiences of emotional/physical pain in that moment aren’t real or to be trusted. In the long term, it is often those who have had their childhood emotions shut down on a regular basis that require psychotherapeutic support, as they no longer quite know how to express how they’re genuinely feeling, perhaps because they don’t really know. And sometimes this can come out physically as well as emotionally. So allowing children the space to express their feelings in those moments of vulnerability and providing empathy rather than judgement (with them feeling validated and heard) can be crucial for ongoing mental health and resilience.
Here are some things we can say instead of ‘stop crying’:
- It’s ok to be sad
- This is really hard for you
- I’m here for you
- Tell me about it
- I hear you
- That was really scary, sad etc
- I will help you work it out
- I’m listening
- I hear that you need space. I want to be here for you. I’ll stay close so you can find me when you’re ready.
- It doesn’t feel fair.
Here a a series of photos of this beauty. We started this journey about four months ago. We’ve done ‘Tox twice now (second time was this week), Dermalfillers about four times now to different areas at each time as to not overwhelm the patient (and to have subtle changes each time). She’s had filler to temples, cheeks, jaw, marionette, nasolabial folds, chin, lip lines and lips. ‘Tox to Forehead,between brows, crows, lips, and chin. Swipe to see progress. Pic one is from this last appointment before we did anything else. Pic Two is side by side of original before and most recent pics (before last round of injections), pic three is before lips and after. Pic four is a series from beginning until now featuring two client selfies (one without makeup to show bruising day after round one and one with makeup day after round one). Slow and steady for this love. This last appointment we refreshed her ‘Tox and added a bit more filler to lips and to some of the lip lines. We will probably do some more filler work in the near future and maybe even ultherapy! .

If you are in emotional discomfort right now you may wish to avoid the feelings. You may want to drink, use drugs, restrict/binge/purge food, self-injure, go shopping, etc. The thing is, you will need to feel those feelings. Yes, it may be uncomfortable. Yes, it may hurt. But just think, if you deal with the feelings now, you will be able to move forward sooner. All feelings have a feeling cycle. All feelings will dissipate over time unless we escalate them. Let the feelings take the natural path. Know that the feelings will increase and peak and then decrease in intensity. Come up with ways to cope during that time. You can do this. Even though it hurts, it’s better for us to feel our feelings than suppress or numb them. 📸:Luc Tribolet

Disclaimer: My legs were absolutely DEAD after this workout lol. My main focus was to increase my volume and work on my form, especially for my hamstrings. For me, those are the most stubborn part of my body, since I’ve pulled my right IT band twice throughout my karate career. I still feel the repercussions from my injuries (tightness/early pull, despite my knowledge of stretching and foam rolling), especially when I do stiff leg RDL’s 😩 My form will be off, but it’s something I’m determined to regain back from the constant stress I’ve put on it from all of the training and competing I’ve done. 😅
I’m gonna do my best to start posting more videos like these for you guys to save for future workout ideas ☺️ Let me know if you guys want to see different videos on here as well!

DIET...... my most hated word ever!
DIET - restrictions, cutting food groups, calories and basically disordered eating.
For more than three decades I was constantly on a DIET... which basically meant ‘will I eat today?’
DIET was restriction. DIET was reducing calories to severe limits. With DIET cane binge eating and bulimia, a f***d metabolism which meant that as soon as I ate I gained weight- we’ve all heard someone say I just need to look at food and I gain weight, well that was me 🙈
With DIET cane excessive exercise to try and limit the weight gain or help the weight loss- tbh not sure which🤷🏻♀️
DIET is a word banded in my life now...
What changed? I was sick and tired quite literally! Injured. And needed to find a healthy me.
I knew what I should be doing- after all I’m a certified Personal Trainer and Nutritionist, but I was too scared to take the first step..... until one night I reached out to a lady online, a lady who has become a good friend. It was a huge step for me, but with that one click my life transformed. I finally took control and started healthy non restrictive eating- no calorie counting, no weighing. I now feel the healthiest I’ve ever felt. So What made the difference?
Being part of an amazing team, all of who are working towards being the healthiest version of them through exercise and clean eating!
